It is a cream ingredient sold as a needle-free way to smooth expression lines. Two small trials disagreed: one showed less wrinkling, one showed none.
Sold for
Argireline is marketed as a needle-free alternative to Botox that relaxes expression lines when applied as a cream or serum.
Studies say
One small randomized trial found a measurable reduction in eye-area wrinkles and another found none, while penetration studies show almost none of the peptide reaches the muscle layer, so the evidence supports a modest surface effect at best (grade B).
Why grade B. For wrinkle reduction there is one randomized placebo-controlled trial in 60 Chinese adults showing a benefit, a 19-person split-face double-blind study showing none, a 26-patient uncontrolled series and the manufacturer's original volunteer study. Skin penetration work in human and animal skin finds very little of the peptide reaches the dermis, which leaves the mechanism in doubt. Small samples and conflicting results keep this at B. 78101214
FDA. Acetyl hexapeptide-8 (Argireline) is a cosmetic ingredient, and the FDA does not approve cosmetic ingredients before sale; products containing it are regulated as cosmetics so long as they claim only to improve appearance. It is not an approved drug for any use. 34

What it is
Acetyl hexapeptide-8 is sold under the trade name Argireline. It is a synthetic peptide of six amino acids (sequence Ac-EEMQRR-amide). It copies the N-terminal end of SNAP-25, a protein that nerve endings use to release neurotransmitters. Lipotec introduced it in 2002 as a cosmetic ingredient. 112
It is one of the most widely used peptides in anti-ageing skincare. It appears in creams and serums at every price. It is sold as a cosmetic, so makers do not have to show the FDA that it works before they market it. 313
How it works
Muscle contraction in the face begins when a nerve releases acetylcholine. That release depends on a set of proteins called the SNARE complex, which includes SNAP-25. Botulinum toxin works by cutting SNAP-25 with an enzyme. Argireline is designed to compete with SNAP-25 for a place in the complex. This destabilises the complex and reduces neurotransmitter release, and nothing gets cut. 1612
In the original lab work, Argireline reduced neurotransmitter release from cells. Its potency was similar to botulinum toxin A, but its maximum effect was much smaller. It also reduced glutamate release from neurons derived from stem cells. 129
The weak point is delivery. The peptide weighs about 889 daltons and carries charges at skin pH. Both make it hard to cross the outer skin layer. Researchers tested it in diffusion cells on human cadaver skin. Most of the applied peptide washed off, 0.22% stayed in the stratum corneum, and about 0.01% reached the epidermis. None was found in the dermis. Facial muscles lie below the dermis. 910
What human research shows
The strongest trial randomized 60 Chinese adults 3:1 to Argireline or placebo. They applied it to crow's feet twice daily for four weeks. Subjective anti-wrinkle benefit was 48.9% in the Argireline group and 0% with placebo. Silicone-replica measurements of skin roughness fell significantly only in the treated group. 7
A 2023 double-blind split-face study in 19 women compared a hyaluronic acid serum with and without Argireline for four weeks. Researchers scored the skin with a Visia camera system. Wrinkle scores fell slightly on both sides. Neither the change within each side nor the difference between sides was statistically significant. 8
The maker's 2002 report described up to 30% less wrinkle depth after 30 days in healthy women using an emulsion of the peptide. A 2020 retrospective series in 26 patients used a 10% gel-cream to camouflage scars. It reported better skin measurements and self-image. Neither study had a control group. 1214
| Study | Model | Finding |
|---|---|---|
| Wang 2013, Am J Clin Dermatol | Humans, n=60, randomized placebo-controlled, 4 weeks | Periorbital wrinkle roughness fell significantly with Argireline and not with placebo; subjective efficacy 48.9% vs 0%. 7 |
| Visia split-face study, 2023 | Humans, n=19 women, double-blind split-face, 4 weeks | Adding Argireline to a hyaluronic acid serum produced no statistically significant wrinkle reduction versus the serum alone. 8 |
| Skin penetration, Kraeling 2015 | Human cadaver and guinea pig skin, in vitro diffusion cells | About 0.01% of applied peptide reached the epidermis and none was detected in the dermis after 24 hours. 10 |
| Blanes-Mira 2002, Int J Cosmet Sci | Cultured cells plus uncontrolled volunteer study | Argireline interfered with SNARE complex formation and reduced wrinkle depth up to 30% over 30 days in volunteers without a placebo arm. 12 |
| CIR safety assessment, 2025 | Expert panel review of toxicology and use data | Safe as used at concentrations up to 0.005%; data insufficient to conclude safety at higher concentrations. 11 |
Safety
In the studies that exist, Argireline is well tolerated. The 60-person randomized trial, the 19-person split-face study and the 26-patient series reported no allergic reactions or irritation. The original developers reported no oral toxicity or primary irritation in animals. 781214
In 2025 the Cosmetic Ingredient Review Expert Panel concluded the ingredient is safe in cosmetics at the concentrations now reported in use, up to 0.005%. It said the data were not enough to judge safety at higher concentrations. Many products and most clinical studies used far higher concentrations, so that gap matters. Chemical analysis of commercial products has also found the peptide partly oxidised at its methionine residue. That raises a shelf-stability question that no clinical study has tested. 51113
Questions for your clinician
- Is there a trial showing this ingredient works at the strength in the product I am looking at?
- Does the evidence say whether it reaches the muscle layer under the skin?
- How does a cream with this peptide compare with treatments that have trial data for wrinkles?
Questions people ask
How does Argireline compare with Botox?
No. Botulinum toxin is an injected enzyme. It cuts SNAP-25 inside nerve endings and paralyses the muscle for months. Argireline is a topical peptide meant to compete with SNAP-25 in the same complex. It cuts nothing and is far weaker. It also has to cross the skin first, and penetration studies suggest it mostly does not. 61012
What do studies say about Argireline and wrinkle reduction?
The evidence is mixed. A 60-person randomized trial found a significant reduction in crow's feet after four weeks. A 19-person split-face study found no significant effect. Any benefit is modest and short-term. No study has followed users beyond a few weeks. 78
Why doesn't Argireline penetrate the skin well?
It is a fairly large molecule that attracts water. It carries both positive and negative charges at skin pH. These properties keep it in the outermost skin layer. In human skin tests none reached the dermis, where the target nerves and muscles sit. 910
Can better formulations fix the penetration problem?
Partly, in the lab. A water-in-oil-in-water multiple emulsion increased penetration into pig skin compared with simple emulsions. Chemically modified lab-made versions also penetrated human skin better in vitro. No one has shown that any of these improve wrinkles in people. 917
