Does Amycretin work?
Partly: early trials of 125 to 186 people, with many dropouts.
The published human data are a first-in-human phase 1 trial of the oral tablet in 144 people, a 125-participant phase 1b/2a trial of the weekly injection, and a 186-participant phase 2 trial of the oral form in type 2 diabetes published in 2026, all randomized and placebo-controlled. In the obesity trials weight change was a secondary or exploratory endpoint and dropout was high, so the weight-loss use is grade B until the phase 3 programme reports. 135
The injection was tested in a phase 1b/2a randomized placebo-controlled study at a single site in San Antonio. In five parts, 125 adults with overweight or obesity were assigned to amycretin or placebo. Treatment lasted up to 36 weeks. Weight loss was significantly greater than placebo in every dose-escalation group, and the effect grew with the dose. The authors noted many people dropped out, mostly for reasons unrelated to side effects. 3
The tablet went through a first-in-human randomized phase 1 trial in 144 people with overweight or obesity. It was also published in The Lancet in 2025. Weight change over 12 weeks was tracked as an exploratory outcome. In 2026 The Lancet published a 36-week phase 2 trial of the tablet, under the name zenagamtide, in 186 adults with type 2 diabetes. HbA1c fell by 0.9 to 1.4 percentage points across the three trial doses, significantly more than with placebo. Stomach and gut effects were the most common adverse events. 15
