Is Setmelanotide safe?
Trial and label data; skin and mood warnings
Skin darkening is the signature side effect. It occurs in most patients, along with darkening of existing moles and new ones. The label advises full-body skin checks because dark lesions are harder to monitor. Nausea, vomiting, reactions where the shot goes in, and headache are common. Spontaneous erections in males and changes in sexual arousal are reported. Bremelanotide shares this class effect. The label warns of depression and suicidal thoughts and recommends monitoring. Some hypothalamic obesity patients also have central diabetes insipidus. In them, sodium imbalances occurred, and acute adrenal insufficiency was reported. The trials treated small numbers for one to a few years. So rare harms and lifelong effects are not known. 123
