Does Triptorelin work?
Yes, in controlled trials, including one of 284 men.
Triptorelin's prostate cancer use rests on randomized comparisons with leuprolide, the reference GnRH agonist, including a 2003 BJU International trial of 284 men showing equivalent maintenance of castrate testosterone over nine months. Hormonal suppression is a surrogate endpoint, but it is the accepted basis for approval of this drug class, so it grades A for its approved use. 12
In the 2003 BJU International trial, 284 men with advanced prostate cancer were randomized to monthly triptorelin or monthly leuprolide for nine months. At day 29, 91 percent of triptorelin patients had reached castrate testosterone, against 99 percent on leuprolide. By day 57 the two were equal (98 percent and 97 percent). Keeping castration from day 29 to day 253 was also equal. Nine-month survival was 97 percent with triptorelin and 90.5 percent with leuprolide. 1
The 6-month depot was approved for prostate cancer. As Triptodur, it was approved for central precocious puberty. The basis was a single-arm open-label study in 44 children aged 2 to 9. In the large majority of treated patients, LH fell to prepubertal levels. 23
